演題: Mechanisms that resolve replication blocking lesions, from crosslinks to G-quadruplexes

要旨:
My group focusses on deciphering the molecular details of processes that
maintain genome integrity. Specifically we are interested in replication fork
blocking lesions such as DNA interstrand crosslinks (ICLs) and G-quadruplexes.
We use a powerful biochemical system to recapitulate DNA replication and
repair under physiological conditions in vitro, based on Xenopus laevis egg
extracts.
I will focus on our latest findings regarding the resolution of mutagenic
G-quadruplex (or G4) structures. While at least 10 different DNA helicases
show G4 unwinding activity, mechanisms of how G4s are resolved in cells are
unknown. We have recently identified a comprehensive G4 unwinding mechanism
that is coupled to DNA replication and involves two helicases, DHX36 and
FANCJ. In addition, I will discuss our most recent results on another,
replication-independent, G4 resolution mechanism.
