演 題:Harnessing charge-mediated immunoreceptor signaling in T cell therapy
要 旨:
T cell immunity is a major pillar of the immune system in fighting with invading pathogens and tumors. Triggering of T cell immunity is orchestrated by a panel of immunoreceptors, including the antigen receptor TCR, the costimulatory/coinhibitory receptors and the cytokine receptors. Most of the immunoreceptors contain a juxtamembrane Basic Residue Rich Sequence (BRS) that can form an electrostatic network with neighboring lipids, proteins, and ions. Using TCR as a system, we show the importance of the BRS signaling in regulating safety, sensitivity, and specificity of T cell immunity and how to tune BRS signaling to promote the efficacy of TCR-T cell therapy. Moreover, introducing the BRS into Chimeric Antigen Receptor (CAR) improves antigen sensitivity and long-term persistence of CAR-T cells. Thus, harnessing natural charge power within the cellular signaling network represents a new
direction to improve therapeutic efficacies of immune cell therapies.
