Molecular mechanisms of nucleosome recognition

要 旨:
We recently reported a nucleosome affinity proteomics screen showing that the nucleosome acidic patch is required for the majority of chromatin interactions. However, the coarse mutation strategy used for our nucleosome library prevented us from defining molecular details of acidic patch binding. We have now completed a second-generation screen to probe acidic patch recognition at amino acid resolution proteome wide. Our screen revealed that the histone H3 lysine 36 (H3K36) JmjC demethylase KDM2A, but not its paralog KDM2B, exhibits a strong acidic patch requirement. We used a covalent conjugate between a JmjC inhibitor and the nucleosome to trap KDM2A and KDM2B for visualization by cryo-electron microscopy (cryo-EM) to understand paralog specific mechanisms of H3K36 demethylation.
