演題: Toward sub-nanometer-resolution analysis of chromatin structure and function

要旨:
High-resolution structure analysis of chromatin protein complexes has been
only feasible with a large quantity of homogeneously purified recombinant
proteins, while many variations, such as DNA sequences, DNA/protein
modifications, and interacting proteins/RNAs, may dynamically alter their
structure and function. To reconstruct high-resolution structures of specific
chromatin protein complexes that are acting on functional chromatin, we devise
Magnetic Isolation and Concentration (MagIC)-cryo-EM, which allows single
particle cryo-EM reconstruction using very small amount of samples, such as
those obtained by chromatin immunoprecipitation (ChIP). The concept and
applications of MagIC-cryo-EM will be discussed.
