演　　題：Mutational Landscapes : from Yeast to Cancers

要　　旨：
The acquisition of mutations, genome rearrangements and ploidy variations play critical roles in adaptation, evolution and tumorigenesis. Whole genome sequencing has allowed extraction of mutational landscape signatures mostly in tumors but it remains difficult to retrospectively determine the mechanistic origin(s) and trajectories of transient or persistent genome variations. Using the model yeast S. cerevisiae, we conducted the reciprocal approach to inactivate single or multiple evolutionary conserved genes involved in distinct genome maintenance processes such as replication, DNA repair, recombination, DNA damage signaling or oxydative stress response. This study revealed the diversity, complexity, and ultimate uniqueness of each mutational landscapes. Several examples of “mutomes”, their trajectories and parallel to tumor-derived signatures will be presented.
